New MidasTM cell line introduced with MON1 chemically competent cells

Monserate announces the first product in our new line of MidasTM competent cells: MON1 chemically competent cells. These high transformation efficiency cells add another useful tool for molecular biology research to Monserate’s growing family of products. Use these cells in conjunction with our low DNA content enzymes such as Taq and Phi29 polymerase exo- Klenow and T4 DNA ligase for better cloning results.

MON1 chemically competent cells are from a new E. coli strain developed in-house for high transformation efficiency (greater than 1 x 109 cfu/µg). MON1 has all the features of popular workhorse chemically competent cells such as DH10B and TOP10. They can be used as a host for most plasmids including large plasmids derived from BAC vectors. These cells are recombination deficient (recA-) and suitable for blue/white selection on X-gal plates utilizing lacZ alpha complementation. They generate high-quality sequencing and plasmid DNA because of an endA- mutation.

MON1 chemically competent cells come in a package with 20 one-shot vials containing 100µl of cells each, 10ml SOC recovery meda and 50µl of 10ng/ml pUC19 supercoiled plasmid DNA as a control. The MON1 genotype is endA1 recA1 mcrA Δ( mrr-hsdRMS-mcrBC ) Φ80lacZΔM15 ΔlacX74 Δ(ara,leu)7697 araΔ139 galU galK nupG rpsL, F-, λ-

Taq Polymerase newest addition to Opposable ThumbTM enzymes

Monserate is pleased to announce the addition of another product to our family of Opposable ThumbTM enzymes: Taq DNA Polymerase. This enzyme is useful for nearly all amplification protocols such as multiplex PCR, qPCR, primer extension, site-directed mutagenesis, microarrays and DNA sequencing. Evolve to a higher quality standard with our new Taq Polymerase-the most recent of our high-quality low DNA content enzymes.

Monserate’s proprietary purification process yields a product that is highly active and virtually free from DNA contamination which can cause spurious PCR products. Our polymerase is isolated from recombinantly-expressed wild type Thermus aquaticus. Taq is a thermostable DNA polymerase with 5’-3’ polymerase activity and an error rate of one per 4.5 x 104 bases. It exhibits deoxynucleotidyl transferase activity, which results in the addition of extra adenines at the 3'-end of PCR products for use with TA cloning formats.

Monserate Biotechnology Group’s high-quality low-DNA content Taq Polymerase is available in both 500 unit (#2001) and 5000 unit (#2003) amounts with a 5 unit/µl concentration as well as a 5000 unit amount with a 50 unit/µl concentration (#2002) which is suitable for high throughput robotic applications. Taq Polymerase is supplied with either 1ml or 3ml of 10x reaction buffer.

Monserate adds Phi29 Polymerase to Opposable ThumbTM enzymes

Monserate Biotechnology Group announces the introduction of Phi29 Polymerase. Φ29 Polymerase joins our existing Opposable ThumbTM enzymes exo- Klenow and T4 DNA Ligase to form a family of low DNA content, high-quality products for molecular biology. The next step in polymerase evolution, Phi29 is able to synthesize greater than 70kb long continuous stretches of DNA and can generate high yields of amplified DNA from small amounts of template. These properties make it ideal for techniques such as Rolling circle amplification (RCA), Multiple displacement amplification (MDA), unbiased amplification of whole genome template preparation for sequencing, protein-primed DNA amplification and in situ genotyping with padlocked probes.

Phi29 is a highly processive polymerase possessing strand displacement capability and 3’-5’ exonuclease (proofreading) activity which is derived from the Bacillus subtilis phage Φ29. It is expressed as a recombinant polypeptide of 66 kDa. As with all of our Opposable ThumbTM enzymes it undergoes our proprietary purification process which virtually eliminates bacterial DNA contamination. Phi29 incorporates nucleotides at a rate of 2000 nucleotides/min. at 30ºC.

Monserate Biotechnology Group’s high-quality low-DNA content Phi29 Polymerase is available in both 1,000 unit (#4001) and 10,000 unit (#4003) amounts with a 10 unit/?l concentration as well as a high concentration 10,000 unit amount with a 100 unit/µl concentration (#4002) which is suitable for high throughput robotic applications. Φ29 Polymerase is supplied with either 1ml or 3ml of 10x reaction buffer. The high concentration is also supplied with 1ml of 1x dilution buffer.
